a- ENZYME XYLANASE ACTIVITY TEST USE DNS METHOD TO KNOWN HIGH ACTIVITIES OF ENZYME XYLANASE

Xylanase is extracellular enzim which can hydrolyze Xylan (hemicellulose) become Xylo-oligosaccharides and Xylose. DNS is reagent that can be use to calculate the quantitative assay of glucose. This reagent is function to give colour to the solution, so that can read by spectrofotometes vis which read the colour from the solution. Quantitative analysis of glucose with Dynitrosalicylic Acid (DNS) method. The reaction with DNS occur are Oxydation and reduction reaction in aldehyde form of glucose and oxidized become carbocsyl fotm. Meanwhile the DNS as oxidizer will reduced and form 3-amyno and 5-nitrosalicylic acid. The reaction goes by alkaline condition. When there are reducing sugar on the sample, so the DNS solution which was initially yellow will reacting with reducing sugar that causing the DNS solution’s colour become reddish orange.
